Introduction
I am Takahito Ayano. I am from Sakai City, Osaka. Currently I live in Fukui, while studying biology at Fukui University graduate school, I am studying the expression regulation mechanism of genes not dependent on DNA sequences, using yeast as a model organism. Experiments are performed while predicting approximate results in experiments, but sometimes unexpected data may appear. In such a case, think about the reason, experiment what to do to prove it, and experiment to prove my own idea. By the repetition, it is a lot of fun to discover and mechanism that does not become yet clear. My hobby is movies and music, drama, playing tennis, games, reading books, and more than anything is that drinking with friends. Especially if you have a good conversation with delicious whiskey, it is the best. I love Godzilla and Gamera. Especially, I will look back at the Heisei's three gamera and VS series many times. If the drama is Gray's Anatomy, the balance between serious and romance is exquisite. In dangerous detectives the multiplication of Hiroshi Tachi and Shibata Kyohei is the best among individuals. I will finish my introduction around here.

Study purpose
There are various kinds of phenomena related to epigenetics, such as cell differentiation, canceration of cells, lifestyle diseases and the like. In these phenomena, chromatin aggregation state changes due to methylation of DNA and posttranslational modification of histone proteins, and gene expression is controlled by a DNA sequence independent mechanism. These regulatory mechanisms are widely preserved in eukaryotes. I study budding yeast as a model organism about this epigenetic expression regulation mechanism. One of the epigenetic expression regulation mechanisms is the silencing mechanism mediated by the Sir protein complex. First of all, as a priori knowledge, a gene is DNA that encodes a protein, and usually the DNA chain is wrapped around a protein called histone and forms chromatin. Next, expression of a gene means that the gene is transcribed, RNA is produced, protein is produced through translation. In this process, since proteins can not be produced unless transcribed, the cells always control transcription, ie expression, according to their own circumstances. This expression regulation mechanism depends on DNA sequence, and it depends on DNA sequence. In the DNA sequence-dependent mechanism, a negative regulatory mechanism that recognizes the sequence of DNA and inhibits transcription binds and suppresses transcription, and a protein that activates transcription binds and positive regulation that activates transcription There is a mechanism. Depending on the change in the cell, these induce the regulation of the protein to which the inducer binds to the DNA sequence, regulate transcription by avoiding the binding substance by changing the initiation point of transcription, transcription including transfer There are a variety of things such as controlling the expression by. On the other hand, in mechanisms independent of DNA sequences, methylation of DNA and posttranslational modification of histone proteins occur due to environmental factors such as GTP changes and stress DNA damage, changes the chromatin aggregation state, Is changing. Among others, in the silencing mechanism mediated by the Sir protein complex, expression of the gene is suppressed in the silencing region in which the histone is deacetylated by the Sir protein, the higher order structure of chromatin is changed, and aggregated state . This region regulates the expression state of the gene by moving as necessary. In order to investigate why this mechanism works and what its biological meaning is, I am analyzing the expression mechanism of specific genes.
